Production process
1. Raw Material Preparation
- Cleaning: Bitter melon raw materials (fresh or dried bitter melon, bitter melon seeds) are initially cleaned with tap water, followed by high-concentration electrolytic ozone water at an ozone concentration of 10-20 mg/L and a water flow rate of 150-450 L/h.
- Pulping and Crushing: The cleaned bitter melon raw materials are then pulped and crushed.
2. Extraction
- Purified Water Addition: 5-10 times the mass of purified water, with the system pH adjusted to near neutral using a buffer salt system, is added to the crushed or pulped bitter melon material.
- Ultrasound-Assisted Countercurrent Extraction: Ultrasound-assisted countercurrent extraction is performed 2-4 times at 38-52°C, with each extraction lasting 30-60 minutes.
3. Centrifugation for Residue Removal
- Centrifugation: A disc stack centrifuge or tubular centrifuge is used to remove impurities, yielding a centrifugal liquid.
4. Microfiltration Membrane Filtration
- Microfiltration Membrane System: The centrifugal liquid is filtered through a microfiltration membrane system with a molecular weight cut-off of 0.1μm to 3μm to remove large molecular impurities, resulting in microfiltration permeate.
- Operating Conditions: The operating temperature is 15-35°C, and the operating pressure is 0.1-0.5 MPa.
5. Ultrafiltration Membrane Purification
- First Ultrafiltration: The microfiltration permeate is filtered through a spiral-wound ultrafiltration membrane purification system with a molecular weight cut-off of 50-200 kDa, yielding ultrafiltration permeate.
- Second Ultrafiltration: The ultrafiltration permeate from the first stage is further filtered through a spiral-wound ultrafiltration membrane purification system with a molecular weight cut-off of 500-1000 Da, resulting in ultrafiltration retentate.
- Operating Conditions: The operating temperature is 15-35°C, and the operating pressure is 0.3-0.8 MPa.
6. High-Pressure Reverse Osmosis Membrane Concentration
- Reverse Osmosis Membrane System: The ultrafiltration retentate is concentrated using a spiral-wound high-pressure reverse osmosis membrane system with a molecular weight cut-off of 100-150 Da to remove water and some residual inorganic salts and small molecular impurities, resulting in a concentrated liquid with a solid content ≥30%.
- Operating Conditions: The operating temperature is 15-35°C, and the operating pressure is 3.5-5.5 MPa.
7. Drying
- Drying Method: The concentrated liquid is dried to obtain bitter melon extract powder with a polypeptide protein content of no less than 25%.
- Drying Technique: Low-temperature spray drying (flash drying) is employed.
8. Enzymatic Hydrolysis
- Enzyme Hydrolysate Preparation: Bitter melon powder is mixed with water, the pH is adjusted to 8-9, and the mixture is soaked for 15-18 hours. Solid-liquid separation is performed to obtain the supernatant. The supernatant's pH is adjusted to 4-6 for precipitation, followed by solid-liquid separation to collect the solid portion. The solid portion is then mixed with water, heated at 85-100°C for 20-30 minutes, and homogenized to obtain a suspension.
- Enzymatic Reaction: The suspension's pH is adjusted to 5-8, and a composite enzyme preparation consisting of flavorzyme, papain, and pepsin is added for enzymatic hydrolysis at a temperature of 45-65°C for 2-6 hours.
9. Enzyme Inactivation
Enzyme Inactivation Conditions: The enzyme hydrolysate undergoes enzyme inactivation treatment at a temperature of 95-125°C for 0.5-2 hours.
10. Purification
- Filtration and Spray Drying: The enzyme-inactivated hydrolysate is centrifuged, and the supernatant is first filtered through a plate and frame filter press, followed by ceramic membrane filtration, yielding active peptide permeate. This permeate is then concentrated and spray-dried to obtain bitter melon active peptide powder.
- Spray Drying Conditions: Spray drying is performed at 80-105°C.
11. Quality Control
- Purity Detection: Bitter melon peptide is purified using ion exchange chromatography, gel filtration chromatography, or other appropriate methods to ensure a polypeptide protein content of no less than 25% and a polypeptide protein molecular weight distribution between 4.5-68 kDa.
- Lyophilization or Spray Drying: The purified bitter melon peptide powder is obtained through lyophilization or spray drying.
